Abstract

Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder of multifactorial etiology, characterized by impairments in communication, social interaction, and restricted behavioral patterns. Given its wide clinical heterogeneity, neurobiological investigations seek to identify underlying brain alterations, highlighting the excitation/inhibition (E/I) imbalance hypothesis. It is suggested that the brains of individuals with ASD exhibit increased excitatory activity or deficient inhibition, a theory supported by the occurrence of seizures in approximately 30% of patients. In this context, calcium-binding proteins emerge as crucial markers due to their role in regulating neuronal homeostasis. Among them, calbindin-D28k, encoded by the CALB1 gene, stands out for its direct role in maintaining the balance between excitatory and inhibitory neurons, serving as an important marker for interneurons. The present study aimed to map the expression of this gene in the Dorsolateral Prefrontal Cortex (DLPFC), an essential region for executive functions and emotional regulation. The research utilized the Allen Human Brain Atlas - Autism Study database, analyzing digital histological samples from post-mortem brains of 22 individuals (aged 2 to 16 years), including both ASD and control groups. Samples were processed using in situ hybridization (ISH) and Nissl staining. Technical analysis involved area delimitation in FIJI-ImageJ software and automated cell quantification using Ilastik software. The results indicated no statistically significant differences in total neuronal density between the groups in the DLPFC. However, ISH analysis revealed an increase in CALB1 gene expression in the ASD group. This finding suggests a possible compensatory response in an attempt to restore neurobiological balance in the face of the disorder's dysfunctions. It is concluded that ASD involves complex alterations in neurogenesis and molecular signaling. Although the lack of differences in neuronal density may be related to limitations such as sample size and anatomical localization specificity, the increased calbindin expression reinforces the importance of investigating genetic biomarkers for a deeper understanding of the pathophysiology of autism.
